Manual of forensic odontology.Fragile X syndrome (FXS) is the most common inherited form of mental retardation, associated with mental and physical malformations and an increased risk for autism and epilepsy. A leading hypothesis for its etiology is that it is due to altered function of the X-chromosome bearing fragile X mental retardation gene (FMR1). Although the basic defect in FXS is thought to be the silencing of FMR1 expression from the X chromosome in the FXS brain, the X chromosome has normal levels of FMR1 in all cells, and the transcriptional silencing of FMR1 expression is likely to be a secondary effect of transcriptional silencing of other genes on the X chromosome. The central hypothesis of this proposal is that normal levels of FMR1 expression on the X chromosome could cause FXS, in some cases, by reducing the protein expression of other genes which have an influence on FMR1 expression in the brain, especially in the frontal lobe. The aims of this proposal are 1) to examine the role of fragile X expression in multiple regions of the developing and adult brain, to identify which brain regions might be most likely to be affected
